  • Optimized for visualization on GelRed®, GelGreen® and SYBR™ Safe™ precast agarose gels (see Product Notes below for more details)
  • Convenient, ready-to-load format
  • Size range: 100 bp to 10 kb (100 bp and 1 kb ladder in one!)
  • Easy to identify reference bands
  • Suitable for 125 to 250 gel lanes

1 kb Plus DNA Ladder for Safe Stains is a pre-mixed, ready-to-load molecular weight marker containing one tracking dye. It has been optimized for use on GelRed®, GelGreen® and SYBR™ Safe™ precast agarose gels.

1 kb Plus DNA Ladder for Safe Stains consists of proprietary plasmids, which are digested to completion with appropriate restriction enzymes to yield 19 bands suitable for use as molecular weight standards for agarose gel electrophoresis. The digested DNA includes fragments ranging from 100 bp to 10 kb. The 0.5, 1.0 and 3.0 kb bands have increased intensity to serve as reference bands.

  • Recommended gel percentage range: 1-1.4%
  • Optimum separation on 1.2%

1 kb Plus DNA Ladder for Safe Stains visualized on a 1.0% TBE agarose gel precast with GelRed.
Mass values are for 0.5 µg/lane.
 Suggested Load: 5 to 10 µl/lane.

Notes
  • Preparation of DNA:
The double-stranded DNA is digested to completion with appropriate restriction enzymes, phenol extracted and equilibrated in storage buffer.
  • Usage recommendation:
We recommend loading 5 µl (0.25 µg) of 1 kb Plus DNA Ladder for Safe Stains per gel lane on GelRed® and GelGreen® precast agarose gels. We recommend loading 10 µl (0.5 µg) of 1 kb Plus DNA Ladder for Safe Stains per gel lane on SYBR® Safe precast agarose gels.
  • Storage Recommendations:
1 kb Plus DNA Ladder for Safe Stains is stable for at least 6 months at 25°C. For long term storage, store at 4°C or -20°C. If stored at -20°C, mix well after thawing. If not well mixed, the load may float out of the wells.
  • Usage Recommendations:
    • The 1 kb Plus DNA Ladder for Safe Stains is not intended for precise quantification of DNA mass but can be used for approximating the mass of DNA in comparably intense samples of similar size.
    • The 1 kb Plus DNA Ladder for Safe Stains might be used on SYBR Green I precast agarose gels but might require further dilution and optimization. We recommend using SYBR Green I as a post-electrophoresis stain for best results.
    • We recommend using SYBR Gold as a post-electrophoresis stain only, as recommended by the dye’s manufacturer.
    • The 1 kb Plus DNA Ladder for Safe Stains is not intended for use on short mini-gels or fast electrophoresis systems. We recommend a minimum of 7 cm of gel length for proper and efficient separation. Best results occur with migration at 5 V/cm (distance between electrodes) for 60 to 90 minutes.
    • Most gel stains run in the opposite direction from that of the migrating DNA. As a result, the bottom of the precast gel might have a lower concentration of stain, which might result in un-uniform staining of the bottom bands. The length of the gel that will contain a lower stain concentration is highly dependent on the migration conditions (gel percentage, voltage, buffer, run time, etc.). This effect can be counteracted by adding some gel stain to the running buffer.
