Unit Definition

One unit is defined as the amount of enzyme required to convert 1 µg of supercoiled plasmid pBR322 DNA to open circular form in 1 hour at 65°C in a total reaction volume of 50 µl.

Notes
  • Use of Nb.BsmI in NEBuffer r1.1 and rCutSmart Buffer may result in cleavage of both DNA strands. Incubation times of more than 4 hours are not recommended.

  • Not sensitive to CpG, dcm, or dam methylation.
  • Nb.BsmI has 100% activity at 37°C, however there is significant increase of star activity when incubated overnight at 37°C.
