Unit Definition

One unit is defined as the amount of enzyme required to digest 1 µg of λ DNA (dam-) in 1 hour at 37°C in a total reaction volume of 50 µl.

Notes
  • MboII produces DNA fragments that have a single-base 3´ extension which are more difficult to ligate than blunt-ended fragments.
  • This enzyme does not benefit from incubations over 1 hour. Visit "Restriction Endonucleases-Survival in a Reaction" for more information.
  • MboII can remain bound to DNA after cutting and alter migration rate of DNA during electrophoresis. To disrupt binding, add SDS to a final concentration of 0.5% or purify DNA before electrophoresis.
  • May exhibit Star Activity in NEBuffer r1.1.
  • Requires two copies of its recognition sequence for cleavage to occur.
  • Requires two or more sites for cleavage. Please review the enzyme list and recommendations in the following table as well as the background information article.
  • Blocked by overlapping dam methylation.
