Heat shock at 42ºC for 10 seconds results in the highest transformation efficiency for T7 Express lysY competent E.coli (NEB #C3010H and NEB #C3010I). Expect approximately 60% loss in transformation efficiency when heat shocking for 80 seconds (see Figure on the main product page).
