Heat shock at 42ºC for 20-30 seconds results in the highest transformation efficiency for NEB 10-beta competent E.coli (NEB #C3019H and NEB #C3019I). Expect approximately 40% loss in transformation efficiency when heat shocking for 80 seconds (see Figure on the main product page).
