Recovery of DNA from 50 bp to 25 kb can be accomplished with the Monarch DNA Cleanup Columns, supplied in both the DNA Gel Extraction Kit and the PCR & DNA Cleanup Kit (5 µg). Efficiency decreases with longer DNA due to tighter binding to the matrix. We observe higher recoveries for DNA between 50 bp-5 kb than for DNA between 5-10 kb. Longer DNA will also be recovered with reduced efficiencies. For optimal recovery of large DNA fragments, please follow the instructions in the manual, which include heating the elution buffer to 50°C prior to elution.
