Monarch DNA Capture beads are provided as a component of the Monarch HMW DNA Extraction Kits (NEB #T3050 and #T3060). The specialized beads are 4 mm in diameter and made from borosilicate glass and have been optimized for capturing high molecular weight DNA in the Monarch HMW DNA Extraction workflows. The use of large glass beads in the HMW DNA extraction workflow enables fast and efficient washing with no drying steps required. Elution from the beads is fast and complete; all or nearly all of the DNA is recovered. DNA eluted from the beads is also easy to dissolve and ready to use in significantly less time than other HMW DNA extraction methods. The beads should be used in conjunction with the Monarch Bead Retainers (NEB #T3004) and Monarch Collection Tubes II (NEB #T2018) for convenient handling and for best results.

The Monarch HMW DNA Extraction Kits enable complete recovery of input DNA with the use of smooth glass beads.



NEB #T3050: 10 μg lambda DNA (~20 μl, NEB #N3011) was combined with Monarch gDNA Nuclei Prep Buffer to bring the total volume to 50 μl. The Cell Protocol (low input) was carried out from Step 4, omitting enzymes and enzyme incubation steps. NEB #T3060: 10 μg lambda DNA (~20 μl, NEB #N3011) was combined with Monarch HMW gDNA Tissue Lysis Buffer to a total volume of 100 μl. The Tissue Protocol (for ‘very low input’ samples) was carried out from Step 8, while omitting enzymes and enzyme incubation steps. DNA concentration of 100 μl eluates from each kit was measured by OD assessment. Average recovery rate and standard deviation were calculated based on 8-fold replicates.

Quality Control Assay
